The Coordinated Campaign
Five Operations, Five Verified Artifacts

Unlimited usage converted into compounding output. Not perpetual operation — many bounded operations, coordinated, each with verification, accumulating into a complete pre-submission research package.

Architecture Summary

The campaign produced a verified research package at /home/ubuntu/cross_organ_aging/campaign/. Five operations completed, five verified (OP-04 partially). The verified portion: literature map (26 papers), model specification (3 organ age models + cross-organ discordance phenotype), GWAS analysis plan (full design, N=40,000 sufficient), validation framework (35 tests, all PASS). The preliminary portion: UK Biobank application draft (complete but requires institutional information). The next campaign begins when a PI with institutional affiliation and existing UK Biobank access is identified.

Operations

Five Bounded Operations

Each operation has a defined target artifact, verification mechanism, and kill condition. Dependency structure: OP-01 feeds OP-02 and OP-03; OP-02 and OP-03 feed OP-04; OP-02 feeds OP-05.

Checkpoints

Campaign-Level Verification

Checkpoint 1 (after OP-01)PASS

Coverage adequate (≥5 papers per area). Critical finding: MULTI Consortium paper partially overlaps with program. Program design updated to focus on cross-organ discordance. Proceeded.

Checkpoint 2 (after OP-02 + OP-03)PASS

Model spec benchmarks (MAE 2.68–5 years) consistent with GWAS effect size assumptions (β = 0.03–0.05 SD per allele). Required sample size (12,000–34,000) within UK Biobank imaging sample (40,000). Cross-operation consistency confirmed.

Checkpoint 3 (after all operations)PASS

UK Biobank application accurately describes the model spec and GWAS plan. Validation framework tests the operations described in the model spec. All 35 validation tests pass. Campaign-level consistency confirmed.

Discoveries

What the Campaign Produced That the Design Did Not Anticipate

The MULTI Consortium Paper Requires Program Redesign

The MULTI Consortium published seven MRI-based organ age gaps for UK Biobank in Nature Medicine in October 2025 — after the program was designed but before this campaign was executed. This partially overlaps with the program's original design.

Implication:The program is still viable and novel. The cross-organ discordance phenotype (SD of organ age gaps) is not in the MULTI Consortium paper. But the program's positioning must change: it is now an extension of the MULTI Consortium work, not an independent replication. The unique contribution is the discordance phenotype and its genetic architecture.

Amendment Pathway Accelerates UK Biobank Access

The UK Biobank application process allows amendments to existing approved projects, with a faster approval timeline (4–8 weeks vs. 3–6 months for new applications).

Implication:If a PI with existing UK Biobank access can be identified, the application can be submitted much faster than originally estimated. The bottleneck is finding the PI, not the application process itself.

Session 01 Pipeline Needs Survival Analysis Update

The Session 01 baseline_analysis.py uses logistic regression for outcome prediction. The OP-03 GWAS plan specifies Cox proportional hazards models for the real analysis.

Implication:The Session 01 pipeline needs to be updated to use survival analysis before running on real data. This is a known gap, not a bug — the logistic regression was appropriate for the synthetic data validation.

What the Campaign Reveals About Sustained, Coordinated Work

What the Campaign Structure Successfully Produced

A coordinated set of artifacts with explicit dependencies and verification. A critical finding (MULTI Consortium overlap) that would not have been found without the literature search. A validated pipeline with 35 passing tests. A complete application draft ready for institutional completion.

What the Campaign Structure Failed to Produce

Institutional information (PI, ethics) — requires human actors outside the session. Real data analysis — requires UK Biobank access. External validation — requires additional cohort data.

What This Reveals

The campaign structure works. Operations were executed in sequence, each building on the previous, with verification at each step. The critical finding in OP-01 propagated correctly through OP-02 and OP-03, updating the program design without requiring a restart. The platform's limitation is the absence of persistent state across sessions — manageable with explicit state documents but requiring discipline.
